I am studying the Pharmacokinetics of a centrally
acting drug that distributes rapidly into the central
nervous system (CNS). The alpha t1/2 is about 10 to 15
min, and the beta t1/2 is about 2 to 3 hours.
I would like to know what are the formulas that relate
plasma concentrations to effect site concentrations
knowing the three constants of the tri-exponential
decline of the drug (alpha, beta, and gamma) knowing
also the other PK parameters. I would like to know
also if the software Kinetica or Siparwin are able
to able to do this type of correlation between the
plasma concentrations, the effect site concentrations
and the effect.
