Isn't it reasonable to treat the embryo and maternal plasma as separate
organs, and to normalize mg drug to tissue volume? The volume of a 4 mm
diameter embryo should be about 25 microliters (or 25 mg assuming a density
approximating that of water). It may be practical to model the yolk sac and
embryo as a single compartment.
I recall some investigators using tritiated water or deuterium to
normalize drug and metabolite concentrations however one of the the caveats
is that you must establish a priori that your drug (or metabolite) is not
labelled during the experimental time course.
