Dear All,
I have the following scenario
1) a set of plasma concentrations data of drug X which follows 3
compartment body model after IV bolus
2) a set of plasma concentrations data of the same drug complexed with
cyclodextrin after intratracheal administration
When using the deconvolution method of WinNonlin 4.0.1, I realised
that there are only two columns named A and ALPHA available, where the
values from IV data should be entered, but there is no place to set
the values for B, C, BETA, e GAMA terms.
What I did was to set as 3 the number of exponentials and 3 lines were
obtanained for each column A and ALPHA, then I set the columns as
ilustrated below. The program run without problem
A ALPHA
Value of A obtained from IV data Value of ALPHA obtained from IV data
Value of B obtained from IV data Value of BETA obtained from IV data
Value of C obtained from IV data Value of GAMA obtained from IV data
I would like to know if my conduct was ok
Sincerely,
Victor

Dear Viktor,
WinNonlin's deconvolution tool supports up to 9 exponentials in the
model. These can be specified as you described, i.e. by selecting the
Settings tab, and then specifying the number of exponentials. Using your
example, the number of exponentials would be 3. Then, you would fill out
the spreadsheet as follows:
A alpha
1 A alpha
2 B beta
3 C gamma
Where A, B, C, alpha, beta, and gamma are the macro constants from your
Unit Impulse Response (UIR) fitting.
Sincerely,
