Step one try to get a separation you can start with the B.P
method of Methanol water 50:50.
If it does not work try other organic solvents. It takes a
while.HPLC also means High patient liquid chromatography.
The peak of the main analyte should come in less than ten minutes
preferably around six minutes. You can vary the mobile phase
composition or flow rate in order to achieve this.
Step two . Select the internal standard. Ideally the peak of the
internal standard should come out before the main analyte. The internal
standard should also help in pushing the main analyte outside similar
to the role of the organic solvents..
Step 3 Optimize all your chromatographic conditions so that you
ultimately get excellent or beautiful peaks..
