I've got some troubles to find MS/MS conditions for a nucleoside
triphosphate.
I can't find suitable MRM conditions.
Is there someone here, who can help me on this topic?
_Best infusion conditions
_Common ionisation mode and polarity for this kind of drug
_Mobile phase for C18 elution, ion-pairing, etc ?

Hello Fabrice,
Analysis nucleoside triphosphate by LC/MS/MS is very challenging and
just a few of teams around the world perform such assays successfully.
Direct assay (i.e.: without the need of dephosphorylation and assay of
the nucleoside itself) of the triphosphate moiety use either ionic
chromatography (Jansen RS et al., RCMS, 2007;21(24):4049-59) or, as
us, an ion-paring agent (Pruvost et al., JMS, 2008 Feb;43(2):224-33).
You will have also to address issues of possible signal decrease along
injection, adsorption or memory effect.
Lastly, I suppose that you will have to assay triphosphate within
cells. You can refer to this paper from our group as a help for
validation process (Becher et al. JMS, 2003 Aug;38(8):879-90.).
