Dear All,
Working in Pharmacology in Veterinary college in India. A colleague
approached me with a problem. Working on a plant extract it was found
that a lower dose was quite effective while a higher dose did not show
any activity, on repeated trials. Any possible explanations!! some
textbooks describe a therapeutic window ( for pure compounds) but did
not try to explain it

The first question that comes to my mind if the extract is
standardized or not. As plants may have various levels of the active
component that you are trying to extract, the plants that were used
for lower dose could be from a different environment (and therefore
have higher level of the active component) than those used for the
higher dose.

Dr. Ramesh,
Is the extract administered orally? How often? Uptake transporter
saturation or inhibition may be involved or metabolizing enzyme
induction. It is possible that the mixture contains compounds that may
impede the uptake or facilitate the metabolism of the active compound.
The active compound may also induce its own metabolism. It would be
difficult to pinpoint the true mechanism without knowing more details
of the extract's composition.
