Respected members,
We have done a 3 period intravenous bolus PK (over 1 minute) crossover
study in dogs at doses of 2, 4 and 6 mg/kg, respectively, for a new
molecule. We have given a wash out period of 1 week between each dosing.
We found siginificant predose concentrations in period 3. How to
analyse the data?
I thought to calculate the elimination rate constant for period 2 and
use that equation to know the contribution of period 2 concentration
at each time point of period 3. Is that right way to do it? or any
other way of doing?
Kindly suggest me.

Hi all
It is understood from your letter, that you found after 1St and 2Nd
doses that the predoses are ZERO(NILL). If this is the case you might
having saturated elimination, then you should look for to make sure
for the following:
1- Eliminations are proportional to the dose(linear PKS with doses).If
so
2- Wait for longer period of time for wash out period to finished.If not
3- Treated it as non-linear and calculate Km and Vmax constant instead
of the elimination rate constant.Best of luck.
