dear all,
in case of calculating extraction efficiency, area of which P&A we
should consider to find out %recovery, means i have processed my
unextracted sample on particular day then may i have to take area of
sample of P&A extracted on same day or i can take area of sample s of
P&A performed on any day?
what i feel is that i have to compare my unextracted sample area to
very previous P&A sample area
please share ur thought --

dear gaurav,
in case of calculating extraction efficiency, area of P&A run on the
same day must ideally be used for calculation of the recovery
exercise. Since a small change in response can be expected everyday
while using an LC-MS/MS system ,if you take the first run P&A as ur
reference and since you calculate recovery on the basis of your areas
obtained,it will not suit your purpose and actually calculated
recovery may not be a true representation of your method.

Dear Gaurav
You should consider extracted samples of P&A that anlysed on the same
day when you prerared unextracted samples.You should not take
extracted samples of P&A processed on any day but if ur P&A analysed
on yesterday i.e on 23-04-08 (last batch), then you perform extraction
recorvy by prepared unextracted on next day i.e 24-04-08 .(first batch)

You can take either area of very previous run or very next run but
where minimum time gap is there so instrument response should not play
any role for example today if you have injected extracted and tomorrow
if you inject un extracted and by chance instrument response is 25 %
high then may play role in calculation.
