dear members,
we work on LC-MS-MS on different molecules where we come
across a common problem in which the response of both the analyte and
IS keeps decreasing irrespective of changing the mobile phase, column
etc., this trend we observe more often in a few molecules. Is there
anything to do with ion competition or something like that. Please
kindly let me know the reason for the problem and please share your
experiance.

Dear naren,
Ion sppression may be responsible for the observed phenomenon. A good
sample cleanup is desired. In your case, phospholipids from the matrix
might have deposited on the column and might have eluted with the
subsequent injections resulting in reduced response. A sample cleanup
using SPE or eluting lipids with a gradient flow may resolve your
issue...
please clarify if i am wrong...

Dear A.SESHADRI.
The one reason for response down is
temperature fluctation of the ac's, Air conditioner temp variation of
2 degree will give u such kind of lower down response. control the
moment in the Mass room to a minimum.
