Dear All, i am facing a problem with a polar basic compound with UPLC.
we are not able to retain the compound on an acquity uplc phenyl /c18
column unless we run with a gradient flow program. 0.1ml/min > 1ml/min
> 0.1ml/min. is there a chance that the first 3 points in calibration
curve fail due to the gradient flow program?

Try a pentafluorophenyl column, possibly with an ion-pairing reagent
such as TFA or HFBA in various concentrations.

Dear all:
For this kind of compound you should use HILIC column type because
this column retains very polar compounds as normal phase but using
aqueous mixes.
For more details, further information will be necessary about this
compound.
