All,
We did not observe conversion of dexamethasone phosphate ester to dexamethasone in rat liver s9 fraction (cryopreserved). I have come across publications where freshly prepared liver s9 fractions have been used for phosphatase activity. I would appreciate it if anyone could share information/experience of evaluating alkaline phosphatase activity with different matrices (fresh vs cryopreserved) from human and preclinical species.
