Dear all
I will measure saliva theophylline level by TDx. But I am not sure
that this instrument has capacity to use for measurment saliva
theophylline level. Has anyone ever used this instrument? Did it work?
I mean that this instrument has appropriate sensitivity? I saw some
papers use this instrument for measuring saliva theophylline level but it
was not mention that about the problem. So I would like to know, wheather
there was a problem or not. Anybody who has an experience for using this
instrument please share the idea.

TDX is a company that makes a lot of different products (ion selective
electrodes, etc.) but I don't see anything on their web site that leads me
to believe there is anything there that will specifically help you measure
theophylline? We monitored theophylline pharmacokinetics in human saliva
by placing an ultrafiltration probe between the cheek and the gum
(absolutely painless and pretty easy to do...except this small teflon tube
hangs out of your mouth for several hours...if the subject wears glasses
you can hang the tube there to get it out of the way). The collected
ultrafiltrates were then analyzed by LCUV (liquid chromatography with UV
detection). The dose was a single, controlled-release Uniphyul tablet
(400 mg-24 hour release) taken orally. No sample cleanup was necessary.
The ultrafiltrates were directly injected into the chromatograph. The
procedure, including the analytical method, is described in Analytical
Chemistry Vol. 64 (1992) p. 2831-2835. I've taught this technique to high
school teachers who are now using it with chemistry students to monitor the
pharmacokinetics of acetaminophen in a chemistry teacher.
