Inuline, extracted from dahlia or burdock roots, or Sinitrin, a
similar polyfrucotsan extracted from algae, were traditionally
measured by a colorimetric assay, which had however a limited
linearity range and various interferences with blood sugars.
An ameliorated enzymatic method has been proposed ten years ago
(Kuehnle HF, Dahl KV, Schmidt FH. Fully enzymatic inulin
determination in small volume samples without deproteinization.
Nephron. 1992;62:104-107).
In our center, R. Séchaud has developed an HPLC method some years ago
(Séchaud R, Décosterd LA, Pechère-Bertschi A, Biollaz J, Kesselring
UW. Determination of the polyfructosan sinistrin in biological fluids
by HPLC with electrochemical detection. J Pharm Biomed Anal.
1996;14:483-490). This method has a satisfactory quantitation range
and sensitivity, which enables precise GFR determination based on the
AUC of the marker (T. Buclin, A. Pechère-Bertschi, R. Séchaud, L.A.
Decosterd, A. Munafo, M. Burnier, J. Biollaz, Sinistrin clearance for
determination of glomerular filtration rate : a reappraisal of
various approaches using a new analytical method. J.Clin.Pharmacol.
