Hello,
I am using a spreadsheet for Loo-Riegelman analysis of IV and transdermal
data. It is working well for the first dose, but for the last dose, the
absorption rate at the first time point seems too high. There is some
accumulation from the last dose and there is usually a lag in absorption, so
there is a drop in plasma concentration from the first through the second or
third time points. Does anyone know whether Loo-Riegelman analysis for
percent absorbed time plots can be used after multiple doses, and if so,
what adjustment is made for the residual concentration from the previous
dose?
Thanks,

Dear colleague
Regarding the use of the Loo-Riegelman method for multiple dose data
I'd like to refer to a paper by Erich Nuesch:
Nuesch EA.
Extension of the Loo-Riegelman method to pharmacokinetic data at steady state.
Eur J Clin Pharmacol. 1990;39(1):55-7.
