Hello all,
We are using a sheep model to examine the extraction of drugs across
various tissues, and want to explore some of the mechanisms involved.
One of the drugs is bound to alpha1-acid glycoprotein in humans at about
80-90%. I looked at the plasma binding in sheep and found ~98% - quite
high- and binding to erythrocytes only accounts for some of this, so we
want to see what proteins are involved using some in vitro binding
experments. I also used a radial immunoassy for human alpha1- acid
glycoprotein, but got no result: either there is no antibody cross-
reactivity or the concentrations are very low. I can buy sheep alpha1-
acid glycoprotein from Sigma no problems, but for the life of me I cant
find any concentration data for this protein (or albumin for that
matter) in sheep. Can anyone help?

Dear David,
we have been using a method to measure AGP in rats which is not a radial
immunoassay method. You can find the description of this method in:
-Imamura H et al. Pharm Res 1994; 11:566-70 (A simple and rapid
fluorimetric determination of alpha-1-acid glycoprotein in serum using
quinaldine red).
We recomend you to use this method to measure the physiologycal AGP
levels in the sheep and then you can use the data you obtain, for your
in vitro studies.
