As patients treated with cyclosporine A are submitted to TDM
(therapeutic
drug monitoring), many of the certified hospital clinical chemistry
laboratories (or clinical pharmacology units) should be able to dose it.

Hi everyone,
I need to measure radioactive tritiated cyclosporin in rat blood.Recent
reports suggest using whole blood for cyclosporin measures since it
redistributes between plasma and erythrocytes according to temperature,
concentration, time...etc. Does anyone know how to avoid the quenching
effect
of the blood red colour? The readings came negative due to the quenching
effect.

You can quench the red color in blood with hydrogen peroxide. Be
careful ... the addition of hydrogen peroxide will bubble and you don't
want to spill your radioactive liquid on the bench. Try it with control
blood to get the hang of it.

I am not sure quenching is a problem with RIA whole blood
determinations of CsA, provided pre-treatment is performed with
methanol to remove drug from erythrocytes, followed by centrifugation,
and analysis of the supernatant. For more details see:
