Dear all,
I know this may be not interest of many here, but does any one know a
good mouse anti-P-glycoprotein antibody?
I am doing western blotting of P-glycoprotein during a kinetic study,
but the antibody I bought did not seem to work. Anybody has experience
running western of such membrane proteins?
Thanks

Dear Jim,
There are two ways I have found to improve the blotting of membrane
proteins. One is to isolate the membranes instead of using a whole
tissue homogenate thus decreasing the interference of cytoplasmic
proteins. The second method to increase Western blot signal is
immunoprecipitation.

You are using a monoclonal antibody which will only recognize native
protein. You have two choices: 1) get a polyclonal antibody or 2)
perform the blotting experiment using native PAGE.
