Hi everyone
I'm just wondering , we've been taught that the concentration of drug
in the plasma is often much lower than on the luminal side of the
membrane.
is this just due to drug distribution into tissues, and presystemic
metabolism ? what other factors are involved ?

this is also due to concentration gradient and to dynamics. The volume
of
fluids in the lumen is small but the body volume is high. At the
absorption
point the "local" concentration in the blood could be quite similar to
the
concentration in the near lumen side. But there is a dynamic dilution
phase
that is due to the blood circulation and to the distribution.
