Dear member,
Currently I am evaluating if the PK parameters of a drug are also
applicable
for children. Can I use the adult clearance, protein binding (mainly
alfa
glycoprotein) etc for predicting exposure levels?
I am interested to learn about your experience in comparing the PK in
children of different age groups and adults. My first interest is with
the
age group of 12-16, but other information will also be much
appreciated. Any
good references are more than welcome.

Hi,
I wouldn't expect the PK parameters for this age group to be
the same as that of adults. The clearance values for these age
groups may not be too far from each other, but then you also have
to note that the volume of distribution is another important
parameter you have to consider for you to have a good prediction of
the exposure.
A good reference to look at for PK parameter prediction is
N.H.G Holford(1996), A size standard for Pharmacokinetics, Clin
Pharmacokinet 30(5):329-332

Regarding your question: Loeckie and co-authors concluded that
physiological differences were most pronounced up to 6 to 12 months of
age.
