Dear all,
I am using winnonlin to simulate concentration-time curves under various
dosing regimen. To do that, I implement the known parameter values
together with the dosing regimen and use the simulation option of
winnonlin. I get the plots as expected but I don't know how to obtain
Total AUC associated with each of these dosing regimen in order to
compare total drug exposure. Does anyone could explain how to get the
cumulative AUC in this setting? Should I use another software?

Hi Chantal,
There is a worksheet is the workbook result where there are all the
predicted concentration data. You can extract them and feed back
Winonlin,
use the NCA option and add them up.
However if you simulate using micro or macroconstants, I would think
that
you have got enough to calculate the AUC corresponding to each dose
regimen
and compare.

Hi All,
I've already replied to Chantal but I thought some of the other
subscribers might be interested that almost all the standard
compartmental methods of Kinetica include AUC as an output as standard.
If you need an additional parameter it is very easy to create a soft
coded method to calculate these as well. All these methods may be
simply added to create a template which may be as sophisticated as you
would like. A demo version of Kinetica 4.2 may be downloaded at
http://www.innaphase.com/support_downloads_kinetica.html
However you will have to contact me for a password to unzip the
package; the software is identical to the commercial package, except
that without a license you will not be able to save or print. NCA,
compartmental and population analyses are possible using the pre-built
templates and models, as well as a graphic designer to build your own.
We also provide packages to assist validation.
