Dear All,
I am currently involved in following up compounds which contain an
carboxylic acid functionality and studying their clearance upon IV
administration. Can anyone help me with respect to devising a screening
strategy e.g. in vitro tests and in vivo species which would give the
best
prediction for the human situation (clearance and T1/2) when the major
mechanism of clearance is non P450 mediated e.g. biliary and/or urinary
excretion of unchanged drug. Any ideas/suggestions or references on
this
topic would be greatly appreciated.

There is a need for a review of human variability for conserved
chemicals of
this kind. This has been done by Renwicks group for glucuronidation
(Dorne
et al, 2001, Food & Chem Toxicol v39 1153-1173). If there are good
reviews
of variation in urine & bile production in man out there then I too
would
interested to discover them,
