To dear GroupsBack to the Top
i am doing some estimation of drugs in plasma.
But i am using the mobile phase of ph 7.0 and my drug is soluble in
only3 organic solvent i.e ethyl acetate , methanol and ACN
but after extraction from plasma in ethyl acetate so much interferences
are there. So how can we avoid this interfernces?
Drug is also acidic in nature
Dear Prashant,Back to the Top
There is no quick answer for your problem. As I mentioned in my earlier
you have to adapt combination of different approaches to avoid
interfearing
components. Mainly, you can do it at two stages:
1. You can avoid them at extraction stage - by adapting suitable
extarction
solevents and techniques as quoted in my earlier response
2. You can avoid them at elution stage- by using suitable column, mobile
phase, and detection modes
If you cannot avoid extracting interfering components, you can avoid
co-eluting with your analyte of interest at stage-2 above and achieve
specificity.
If nothing works, shift to MS mode.
Hope this helps.

Dear prashanthBack to the Top
you can try diffrent type of extraction solvent like dichloromethane,
diethyl ether, t-BME, Hexane and there combinations to remove the
interferences. it may help you.

Dear Prashanth,Back to the Top
You can also try extraction first with relatively non polar solvent
which may remove interfering material and the extract with polar solvent to get
compound of interest OR if the compound is of acidic or basic in nature a double
extraction using acid or base may also help. Hope this solves your problem
