Hello all,
I would like to dose one of our compounds as multiple doses. I wanted
suggestion regarding suitable preclinical formulation to characterize
the
pk and pd.
Few points:
(1) The compound is not soluble in aqueous solution.
(2) Change in PH, the compound is soluble only to small extents, at
slightly
higher doses it will crash.
(3) Cosolvent (PEG 400-chronic administration leads to diarrhea, DMA-i
found in one of the pharmapk archives that it is toxic to mice species).
Given these i wanted to know what will be the generic/safe/simple
formulation for multiple doses for preclinical studies. Any
comments/suggestions are welcome.

Dear Suresh,
Could you tell us what is the route of administration? There are many
cosolvents available for dissolving water insoluble compounds.
Combination of surfactants and cosolvents works the best. You can try
propylene glycol as cosolvent.
