I will highly appreciate if you could provide me the detail informationBack to the Top
regarding the following:
1. While performing some isolated tissue experiments e.g. Guinea pig
trachea, I am finding it difficult to obtain the PA2 value, the one
which is reported in the literature and requires higher concentration
of antagonist than reported one. What is the reason and what othe
opetion I should try to overcome it ?
2. If I incubate the tissue preparation once or 3-4 times (to the other
tissue preparation) with antagonist , (after providing sufficient time
to wash out the antagonist solution in case of 3-4 time incubated
tissue preparation so as to avoid its accumulation in the tissue-bath),
the percent inhibitory response of antagonist is higher with repeatedly
incubated tissue preparation as compared to once-incubated tissue
preparation, against contractile response of the agonist. Whether this
method of frequent incubation with antagonist is valid ?
2. I will again appreciate if you could focus on the inter-relation
between IC50 and PA2, if any.
