Dear all
I am trying to equilibrate the mobile phase, ammonium acetate buffer 10
mM/acetonitrile (95:5 v:v) in the Hplc system but the zero over lamp
lights up and the zero adjustment is not affected. When I switch this
mobile phase to methanol: H2O 50:50, the zero adjustment is working.
Could you tell me what I have to do to solve this problem.

Dear Sayer,
Try using potassium phosphate or sodium phosphate buffer instead
of ammonium acetate
buffer in your mobile phase. I think this will work.

Dear Sayer
Pl check the detector setting in your HPLC method. Normally, Detector
on/off is not related to mobile phase.
