If your IS is giving the same signal, but your analyte is not giving a
consistent or proportional signal look at making a standard curve in
the mobile phase of the analytical system to see if it is a compound
effect or it is rightly so an effect of the extraction procedure. Make
sure that the concentration range that you are working in is consistent
with the standard curve that you propose to generate. Working out
extraction efficiencies and their effects on the analysis is a key part
of bioanalysis and determining absolute response in the mobile phase
will give you more insight into the characteristics of your analyte.
