Sir , here I want to know how to calculate that whether the test
drug is bio-equivalent to reference drug.
How to calculate it by numerically without Winnoline. Please explain
me with example.

go for a book, e.g. Chow and Liu's "Design and Analysis of
Bioavailability and Bioequivalence Studies":

It contains an abundance of numerical examples.
To calculate an ANOVA, M$-Excel will do the job - although I would
not recommend it -, or even with a little patience a pocket
calculator ;-)
To get the confidence interval a table of Student's t is also a
necessity.
