Dear all,
I know I am asking an obvious thing for most of you but, we are
experiencing
several problems in filtering a buffered solution of a lipophilic drug
containing a 0.2% DMSO percentage with a cellulose acetate filter. We
have a
very low recovery. Now, I am thinking it is due to the small percentage
of
DMSO in the solution, which isn't tolerated by filter material. I am
thinking to use PTFE filters of the same porosity. Has anyone ever
experienced troubles with the same filters and similar solutions?
The chief thing is that I should use either cellulose acetate or PTFE
sterile filters to sterilize the drug solution for subsequent cell
colture
procedure. Do you know if using cellulose acetate or PTFE is
indifferent?

Is the filtration a few milliliters by a syringe into a vial or
container for analysis?
Or is filtration of a large volume of solution through a filter?

It is concerning the filtration of a few milliliters solution into the
vial. And I had the same problems also with Nylon filters.
