Hi,
I am studying the tissue distribution of a drug using enzyme assay.
The substrate being used is C14 labeled and I am assessing the
radioactivity present in the product after the enzyme reaction.For
determination of the unknown concentration of inhibitor drug present
in various tissue extracts, I am using standard equation Vi/V0 =
IC50/ IC50 +[C] . The calibration curve (% radioactivity remained vs
concentration ) drawn is different for different tissues. The point
of confusion is that I am getting different IC50 values for each
tissue (drawn from calibration curve ) due to difference in recovery
from different matrices. Can I go for calculating concentrations
using different IC 50 values for different matrices or else shall I
try for some another curve fitting equation? Also how to take the
standard error of IC 50 value in account during calculations?
