Hi,
I am planning to perform intestinal perfusion experiments in rats
using penetration enhancers. I want to measure increase in absorption
enhancement as well as changes in morphology of the intestine. I have
following questions.
1. What perfusion rate is optimum? In the literature, people have
used rates from 0.1 ml/min to 3 ml/min. When I tried to perfuse the
entire small intestine with 0.1 ml/min rate, the intestine became
distended. Does this affect permeability/integrity of the intestinal
mucosa?
2. If I take blood samples while perfusing the intestine, is there a
way to correlate these results to oral absorption?
