Dear All,
Could anyone provide some physical-chemistry information about
prednisone: Log P and/or cLog P and solubility (water).
Informations about permeability assay will be great too.
I would like to know about provisional BCS classification for this
substance.
Regards,

Daniel,
The calculated log P of prednisone is 1.471. The PubChem Compounds
database in the National Library of Medicine lists cLogPs for almost any
compound http://www.ncbi.nlm.nih.gov/. The references list its water
solubility as very slightly soluble which is, unfortunately, not very
informative. Alcohol solubility is 1 gram in 150 mL. Does anyone know
of a
good database that lists solubilities of drugs?

I don't have data nearby for prednisone, but the very close structural
analog dexamethasone is claimed to be 100 mg/L (which seems a little
high to
me). I'm sure prednisone is in that ballpark.

Dear Shelley,
Solubility database: Very informative paper of E. Rytting
et al on Aqueous and cosolvent solubility data of drug like
organic compounds was recently published in The AAPS
Journal 2005, 7(1) Article 10 ((http://www.aapsj.org).
Intrinsic aqueous solubility values of 321 drug compounds
collected fom the literature, as well experimental
cosolvent solubility data for 122 drugs in PEG 400-water
system are listed.
