Dear Colleagues:
Dose any one have a comparative PK data in Rhesus and
Cynomolgus monkeys or can share his/her experience as
which one more closely mimic/predict human PK? Beside
size and price, are there any other reasons one may
choose Rhesus vs. cynomolgus?

Hi Rostam,
I can only comment in practical terms. The use of cynomolgus monkeys
was originally driven by the closing of monkey exports from India in the
1990's; India exported pretty much all of the rhesus monkeys that were
being used. Consequently, it became very difficult to obtain rhesus
monkeys and the focus switched to using cynomolgus monkeys. The
situation is rapidly reversing now, with the supply of rhesus monkeys
increasing and the demand for cynomolgus monkeys outreaching supply.
We currently use cynomolgus monkeys primarily because we do not want to
switch between cynomolgus and rhesus mid-stream in the development
program and because the cost of drug is so high that the generally
smaller cynomolgus monkeys are beneficial.

I do not have comparative data but my contribution would be to say that
I would
be amazed if one was a better model than the other, for human PK,
across the a
diverse range of compounds.
Taking that thought further which human PK are you trying to predict
.... is it
Caucasian, Japanese ... is it people of Japanese stock that are second
generation American... are you talking of children or frail elderly ...
obese or
even ..... ill people ?
Which human is a good predictor/mimic of human PK ?

Jeff and Dave, thanks for your comments. I guess, Tox
labs may have done bridging studies to compare Rhesus
and Cyno as for some programs switched from one strain
to the other may have happened half way through the
program. Would have been nice if more people shared
their experience with the group.
