Dear colleagues,
I plan a lung tissue kinetics in rat because I don't see any plasma
levels
of my administered compound (intratracheal dose, powder, 1 mg/kg).
Stability after an iv dose is acceptable. The question is: How can I get
useful values? Is it necessary to carry out a lavage (BAL)? What else I
have to consider? I would be happy to get proposals and useful
literature.

What is the intention of performing lung tissue kinetics? Is it
metabolism? or you would like to know the actual tissue concentration
in lung tissue? Could you please put your Q with more clarity.

Dear Thomas:
What was the last sampling time points in your
original study? Do you still have the frozen
carcasses? If you believe most of your compound went
to the lung, then you probably can detect it even at
late time points. So as a POC study you can use the
old tissues if they are properly preserved. The other
question is that what do you know about the toxicity
of this compound? Did you observed anything (e.g.,
shortness of breath) during the first PK study?

Sorry for the delay. There was a problem with my email. Thus, I did
not get
your replies.
The reason for performing a lung kinetics is to know the actual
tissue concentration. It would be for instance interesting to
distinguish between not dissolved compound in the lung and the
amount
of dissolved compound that is already in the tissue. At the
moment,
we are not interested in metabolism.
The last time point in our original it study (cold, no 14C)
was 24 h
post dose and there was no measurable plasma concentration
(following
a single intratracheal dose).
Until now we have seen no obvious toxicity of this compound.
