Dear all,
We are getting quite high inter- but also intra-animal variability in
plasma obtained with EDTA as the anticoagulant.
ALP data are much tighter and I am wondering whether it is (local)
tissue damage that could be the cause since we collect through the
retro-orbital sinus. Haemolysis is negligible (by eye).
Does anyone have experience with this?

At first the values of ALT and AST in mouse is obvious little bit
higher. have you generate intera lab data for biochemical parameters?
if yes and then also your values are higher then that values then you
can concentrate on two points. one is anticoagulant, people use
Heparin for biochemical analysis and EDTA for Haematology analysis
because in EDTA the platelets are much stable. and other is your drug
is really generate any adverse effect in liver but only on the basis
of biochemical analysis result you can't conclude this and therefore
you have to go for microscopic analysis of also. And if you can find
any microscopic findings then you can conclude it as an adverse
effect. As far as these two enzymes are concerned, commonly they
increase in relation to other parameters also i.e. Total Bilirubin.
So mainly three points should be cleared from your side.
1. Generation of in-house biochemical normal range.
2. Try to change anticoagulant.
3. Go for microscopic analysis also for that particular mouse liver.
I hope this will solve your problem.
