I would like to find out if anyone has developed an accurate HPLC
method for the quantitation of gentamicin. I have developed a HPLC
method with ELS detection (no derivatization needed) that separates
the different components (C1, C1a, C2, C2b, and C2a); but, I have to
standardize against the USP gentamicin reference standard which has a
given purity in the microbiology assay units of microgram/mg. This
is purity based on activity, not chemical molecule purity.
Therefore, the accuracy validation may or may not pass, depending on
the gentamicin raw material assayed against the USP standard.
Any advice would be greatly appreciated.

That is the basic problem with chemical quantitation of gentamicin--lack
of pure reference standards for the individual components. When I last
checked, USP did not have them. Stefan Soback purified his own in his
2000 publication. There is a remote chance Riedel-de Haen carries them
in the EU, but I didn't see them in Sigma's Vetranal section. Good
