How to determine the degraded compound A 21 desamido insulin in
serum? Do any one has the experience of quantitation of A 21 desamido
insulin along with insulin in serum and
to plot of non- compartment profiling of Insulin using PK software.

dear sir
what i feel is if matrix is different surely the amount will be
vary.If the drug is binding to blood cells the high level need to
be expected .but incase of insulin of the observation may different.
sir this normal biochem books all they are writing only Serum
containing insulin.no were plasma insulin.
I read some paper desamido formed by the mild heat but no strong acid
or alkali. or expired insulin we can able check .

Hi All,
Due to insulin degradation enzyme (IDE) released mainly from red blood
cells, we have seen the recoveries of both endogenous and exogenous
insulin were significantly decreased in serum after freeze/thaw and
bench top as well as in hemolyzed serum or plasma samples. EDTA as a
protenase inhibitor (PI) plus Bacitracin as a PI too, significantly
increased stability of insulin in normal and hemolysed plasma. BAC is a
strong IDE inhibitor that can be used for preventing insulin degradation
during sample preparation, storage and analysis.
