Dear all
I have been trying to develop bioanalytical method for Migilitol.
This compound is highly polar i am getting only 20-30% recovery in
dog plasma. Does anyone have a idea or thought to increase the
recovery. i appreciate all the comments.

Hi,
Maybe you do not need to extract such a highly polar compound. If you
can clean most of the plasma "junk" with the likes of acetonitrile
and DMSO and prepare your standards in the same matrix (dog plasma)
while using a suitable internal standard ( e.g. gabapentin) and try
different compositions of mobile phases (maybe two different
separation columns too). Assay development can be a real hectic process!
