I would like to know the opinion of yours about the recover assay. If
you lose volume, for example, when you put 1ml and recover 0,75ml, is
it correct to express the recover by thinking about how much you
would have if you had recuperated 1ml?

Generally, concentration is the parameter and you might not need to
adjust since concentration is the factor. If you were concerned and
needed a control you could add a compound similar to the analyte
chemically but without activity.
Volume concerns revolve around washing or rinsing and adjusting for a
loss or an increase in volume. Nasal washes and bronchialveolar
generally have a dye added. If the volume is added, the sample has less
absorbance, this would not work for volume loss.

Kelen, it depends by the sd of the recovery, if it is really small
this means that the process is repetible and you can correct for the
recovery, but if the sd is high this mean that procerss is out of
control and you can't.
In any case I suggest you to adopt the Internal Standard technique in
order to avoid any possible mistake.
