Dear members,
As per the analysis requirement drug is to be estimated in whole
blood. Can any body explain in what vacutainers the whole blood is to
be collected and how to be stored.
regards,
kiran

There are examples in the literature of binding of drug to caps of
vacutainers. You must do a control experiment to make sure there is no
sticking to vacutainers.

Dear Kiran,
As Stan Cotler has noted, you'll need to check to ensure that your
analyte does not adhere to the tube or its cap. For whole blood
assays the tube will need to contain an anticoagulant to prevent
clotting. Typical anticoagulants are heparin and EDTA; there are
other less common ones as well. Tubes sometimes also contain a
preservative for the anticoagulant. Since this is for a whole-blood
assay, you will not be using the type of tubes that include a
material to aid in separation of cells and plasma. If you are using
a known or published method, consider contacting the authors for the
specific type of tube they used. In your case, you will need to
ensure that he anticoagulant and any preservative do not interfere
with the analytical method that you use in your lab.

Dear Kiran
As per my knowledge blood samples are collected in EDTA vacutainers,
if the drug is light sensitive, collect in low light conditions,or if
the drug is prone to degrade rapidly, add preservatives like
carbonate buffer (depends upon the nature of active ingradient).
After centrifuging the blood samples plasma samples are to be
preserved under either -15*C/-50*C. The above specifications are for
human blood preocessing for pharmacokinetic analysis.
