I would appreciate all thoughts/ideas on how one would standardize
Cmax and AUC for body weight.
My inclination/assumption is that in general, Cmax and AUC are
inversely proportional to body weight.
All patients have been dosed at the same level (i.e., 10 mg). We now
want to standardize Cmax and AUC for body weight prior to statistical
analyses.
All comments much appreciated.
Thanks,
Jack

Jack,
The only principle based method I know of is to use a compartmental
model parameterised in terms of clearance and volume of distribution.
From basic allometric theory (see http://www.cognigencorp.com/
nonmem/nm/99oct042001.html) one can scale clearance to the power 3/4
and weight to the power 1. This will then account for size related
differences affecting disposition. Cmax will depend on the model
chosen for drug input but this model is unlikely to be size dependent
in the adult age range.
