Can anyone tell me what dose of Midazolam is used in oral
DDI studies? Has to be a therapeutic dose (7.5 mg) or could be lower
(e.g., 2 mg)?

Dear Rostam,
I think it depends on the basic purpose of your study. In literature
the doses from 2-15 mg have been used. However, giving a therapeutic
dose seems to be a good option always.

you get indications of no to borderline interactions with 2 mg, can
you conclude that 10 mg would be the same?

Rostam,
I dont think that you can conclude like that. 10 mg of midazolam may
have effect of different intensity on PgP (inhibition) or CYPs
(induction) as compared to 2 mg dose (I am unaware if somebody has
done this study). So it again may depend on exactly what you want to
study using midazolam.
