use the BP standard start with methanol water 50:50 see whether the
peaks are coming out
Try other organic solvents like acetronitrile and see what happens.
Instead of water try phosphate buffer and see what happens. The
bottom line is that the peaks must come before optimization or
accuracy, linearity, and precision determination.

Dear Senthil,
Based on the pKa of the drug, choose a buffer that will keep your
drug mostly in a single state (either ionised or unionised). This
helps in obtaining sharp peaks due to uniform partitioning. If you
are planning to use fluorimetry, Fluoresence of compounds is also
dependent on the ionised state, so take that into consideration for
choosing a buffer.
