All,
Does anyone know the appropriate PK model for maternal-placental-
fetal transfer? I have a cpd that have transplacental transfer from
mother to fetus. In addition, metabolites of the cpd were detected in
the fetal circulation, indicating metabolism in the placental and/or
the fetal compartment. Any comments are welcomed.
Thanks,
Yan

You may find a good example of PBPK model for transplacental
transfer, which I expanded for lactational transfer, in the following
publications and the references therein:
Clewell HJ, Gearhart JM, Gentry PR, Covington TR, VanLandingham CB,
Crump KS, Shipp AM. Evaluation of the uncertainty in an oral
reference dose
for methylmercury due to interindividual variability in
pharmacokinetics. Risk
Analysis 1999; 19 : 547-558.
Byczkowski, J.Z., and Lipscomb, J.C.: Physiologically Based
Pharmacokinetic Modeling of the
Lactational Transfer of Methylmercury. Risk Anal. 21: 869-882, 2001.
Hope it will help you.
Best wishes.
