Dear all,
Can any one suggest that the pharamacokinetic data of mice can be
extrapolated to Rat pharamacokinetics or vise versa using allometric
scaling. please do the reply with refrences

Dear Dev,
If your compound is metabolised, I would say that you have no chance.
Which mouse? Which rat? Strain, sex, supplier, age can all have
effects as large as species differences, and probably diet and stress
status as well. I am amazed that anyone still practices inter-species
allometric scaling on metabolised drugs. It defies everything we know
about enzymology.
If your compound is excreted renally, then you probably have a good
chance. For biologicals, my experience is that PK parameters are
similar for rat and mouse, surprising for the body weight difference,
and I cannot tell you why, so I would never assume that it is a
mathematical law.
I look forward to reading some stout defence of inter-species scaling!
