Hello,
For PK parameter determination is their any ideal doses (for both iv
and po)? Is one to five mg/kg, iv and one to ten mg/kg , po is OK? Is
the dtermination of PK parameters inculding bioavialability is
correct, if one uses higher doses, say 10 mg/kg iv and 20 mg/kg po?
We found huge differences in bioavailability when po dose is changed
from 5 to 20 mg/kg. Which dose will give the correct bioavailabilty?
Appreciate some good valid comments.

Dear Jose,
You can administer different doses as mentioned in the
query but if the dose linearity was applicable for dose range you have
been administering then the bioavailability also will be increasing in a
linear fashion. Dose linearity plays an important role. Correct dose can
be concluded by conducting exposure studies on trial and error basis.
