DMSO is the preferred acceptable solvent -- I have seen cytotoxicity
with even low concentrations of ethanol. I never had a need to go above
0.1% final concentration DMSO (testing at 10 micromolar final). I think
up to 1% would be O.K., but much above that you start to fluidize the
membrane and loosen tight junctions (so I've read).

Dear Sachin,
I used to use HBSS in Caco-2 study, final organic solvent (we use
DMSO) concentration should be less than 0.5%.
