Dear colleagues,
If some one has an hands on experience in Analyzing and Quantifying
Analytes in Tumor Tissues on LC-MS/MS, Can suggest me how to
approach for sample Prep. Like Extraction procedures, Homogenization
Procedures.

Hi,
The tumours can be simply be homogenized in phosphate buffered saline
(PBS) using a tissue homogenizer. Normally the volume of PBS used is 1:4
(i.e. for 1 gram of tissue use 4 ml of PBS). The homogenate can then be
treated like any other biological matrix. Alternatively, you can even
use Bio Pulverisation technique It is described in this paper "Patel K,
Lewiston D, Gu Y, Hicks KO, Wilson WR. Analysis of the hypoxia-activated
dinitrobenzamide mustard phosphate pre-prodrug PR-104 and its alcohol
metabolite PR-104A in plasma and tissues by liquid chromatography-mass
spectrometry. Journal of Chromatography B 2007;856(1-2):302-311". You
can then either follow a protein precipitation or liquid-liquid
extraction approach for extraction of your analyte and analyse by LC-MS.
Hope this answers your question.
