Dear All,
Now we have performing method development for Adefovir, while
processing freshly spiked sample we were getting good response, the
same samples stored in Bench top or it in deep freezer we unable to
get the response its all most nill,its may be degraded in plasma, can
any one suggest me good stabilization reagent for adefovir in plasma.

What you are spiking in plasma, its Adefovir or Adefovir
dipivoxil and what you are looking while quantitation.

Look at the structure of the molecule. What part of the molecule is

susceptible to degradation by plasma enzymes? Do a search on NLM to
find
how most samples have been collected and what preservatives have been
added
to minimize loss in plasma.

Dear Gopi,
You can try following possibilities:
1) Adding buffer either acidic or basic pH in the plasma containing
drug and then proceed for the processing of the samples. One of our
NCE molecule was stabilized using acidic buffer. This may help in your
case also.
2) Add some plasma esterase inhibitor like dichlorvos in the plasma.

Please check the stability of drug in plasma at different time
interval and also at different temperature.
By doing this you can find out the stability of Adefovir.
dichlorvos is good stabilizing agent.
