We appreciate your help on the % dose calculation in bile.
We have dose 30 uM/Kg compound to the bile duct cannulated rats and
collected bile hourly up to 1 hr post dose. We have received the
concentration in ng/mL. We need to caliculate % dose excreted in bile
at each hour. The experimental conditions and values are as mentioned
below.
Experimental conditions:
Dose: 30 uM/kg
Route of administration: Intravenous.
Rat Bw: 250 g (we have dose 7.5 uM/ rat)
Time point: 0 - 1 hr
Results:
0 - 1 hr bile volume was 1.25 ml and concentration in that were 2000
ng/mL andMolecular weight of the compound was 400. Therefore we
considered 400 ug = 1 uM.
By using these values we have calculated % dose as follows.
Total administered dose to one rat 7.5 uM (7.5 uM x 400 m.wt = 3000
ug/ rat)..Concentration of test compound in 1.25 mL of bile was 2500
ng (2.5 ug) and percent dose excreted between 0 - 1 hr was calculated
as 2.5/3000 x 100 = 0.0833 %.
So test compound % dose eliminated was 0.0833 %.
So, please suggest this is the correct way of calculating the % dose
eliminated in bile.

did you collect BIle for longer than 1 hour?
Did you collect plasma samples? and if so for how long
What was the plasma profile in non-cannulated rats?
