Hi
I plan to do Caco using the BD Biocoat HTS Caco-2 Assay system, does
this shorter time method give similar results to what we get with 21
day Millipore system.
Please help.
Thanks,
Maninder

Dear Maninder,
some years ago, when I tried to speed-up CaCo-2 assays by using
systems able to generate rapidly the sealed monolayer, I was worried
that monolayer architecture can change by changing coating of inserts
(at that time I was using TransWell inserts with different coating
conditions)
This occurrence seemed mainly related to tight-junctions development
and to establishment of correct cellular network, even if trans-
epithelial resistance usually reached the required value (hundreds of
mOhm).
We experienced also a difference on behaviour of trans-epithelial
permeability between slow and rapid growth systems for some compounds,
possibly mainly related to compound-induced changes on intercellular
junctions acting in different way on rapid and slow growth systems.
Kind regards
Stefano
